Phytoplasma Identifi cation in Opuntia monacantha var. variegata from a Private Collection of Russia

Opuntia monacantha commonly known as prickly pear cactus from a private collection of the Samara region, had an unusual shape and color of cladodes, which suggested the presence of infection with phytoplasma that was confirmed using polymerase chain reaction (PCR) in 2015. However, the taxonomic belonging of phytoplasma to the group was not determined due to the low concentration of the pathogen. The prickly pears analyzed were rooted and continued to grow in the greenhouse for five years. As a result, numerous small cladodes were formed on each of the main stems. PCR and analysis of restriction fragment length polymorphism revealed a high degree of infection of prickly pear with witch’s broom peanut phytoplasma ‒ 16SrII group. Restriction patterns obtained after PCR product digestions with AluI, Bsh1236I (BstU1), HinfI, HhaI, HpaII, RsaI, TaqI and TruI1 (MseI) endonucleases allow phytoplasma to be identified as belonging to the 16SrII-С subgroup.
