Идентификация фитоплазмы опунции одноколючковой вариегатной из частной коллекции России

Опунция одноколючковая Opuntia monacantha var. variegata Haworth. из частной коллекции Самарской обл. имела необычную форму и окраску кладодиев, что позволило предположить наличие инфицированности фитоплазмой. Это было подтверждено с помощью полимеразной цепной реакции (ПЦР) в 2015 г. Однако таксономическая принадлежность фитоплазмы к группе не была определена из-за низкой концентрации патогена. Взятые для анализа кладодии опунции были укоренены и в течение пяти лет продолжали расти в теплице. В результате на каждом из основных стеблей, образовавшихся из укорененных кладодиев, сформировались многочисленные мелкие кладодии. ПЦР и анализ полиморфизма длины рестрикционных фрагментов позволили выявить высокую степень инфицированности опунции фитоплазмой группы ведьминой метлы арахиса ‒ 16SrII. Электрофореграммы рестриктов, полученных с использованием эндонуклеаз AluI, Bsh1236I (BstU1), HinfI, HhaI, HpaII, RsaI, TaqI и TruI1 (MseI), позволяют идентифицировать выявленную фитоплазму как принадлежащую к подгруппе 16SrII-С.
